Deletion of the p85 Regulatory Subunit of Phosphoinositide 3-Kinase in Cone Photoreceptor Cells Results in Cone Photoreceptor Degeneration

چکیده
PURPOSE. Downregulation of the retinal insulin/mTOR pathway in mouse models of retinitis pigmentosa is linked to cone cell death, which can be delayed by systemic administration of insulin. A classic survival kinase linking extracellular trophic/growth factors with intracellular antiapoptotic pathways is phosphoinositide 3-kinase (PI3K), which the authors have shown to protect rod photoreceptors from stress-induced cell death. The role of PI3K in cones was studied by conditional deletion of its p85 regulatory subunit.

متن کاملMorphology of retinal photoreceptor layer in continuous light-exposed and dark-adapted male cats
The morphology of retinal photoreceptor layer was studied in continuous light-exposed and dark-adapteddomestic male cats (Felis catus). The eyes of 12 healthy adult cats (4 in continuous light-exposed group, 4 in continuous dark-adapted group, and 4 in control group) were routinely fixed and studied by electron microscope. Results showed that the general structure of photoreceptor layer in this...

PURPOSE To examine the effect of loss of cone photoreceptor cells on retinal degeneration. METHODS We previously identified a cone photoreceptor cell-specific promoter of human cone transducin a-subunit (GNAT2) gene. In this report, a minigene, Trc-Tox176, that contains the GNAT2 promoter, an attenuated diphtheria toxin A-chain gene, and an enhancer element from human interphotoreceptor retin...
متن کاملActivation of oncogenic tyrosine kinase signaling promotes insulin receptor-mediated cone photoreceptor survival
In humans, daylight vision is primarily mediated by cone photoreceptors. These cells die in age-related retinal degenerations. Prolonging the life of cones for even one decade would have an enormous beneficial effect on usable vision in an aging population. Photoreceptors are postmitotic, but shed 10% of their outer segments daily, and must synthesize the membrane and protein equivalent of a pr...
متن کاملA Hybrid Photoreceptor Expressing Both Rod and Cone Genes in a Mouse Model of Enhanced S-Cone Syndrome
Rod and cone photoreceptors subserve vision under dim and bright light conditions, respectively. The differences in their function are thought to stem from their different gene expression patterns, morphologies, and synaptic connectivities. In this study, we have examined the photoreceptor cells of the retinal degeneration 7(rd7) mutant mouse, a model for the human enhanced S-cone syndrome (ESC...
